汇报提要:
肿瘤的发生与演进发源于遗传和表观遗传的异常变动。这些变异导致癌基因的激活、抑癌基因的失活、以及其他分子和信号通路的变动,使正常细胞发生转化、癌细胞进一步恶化。发现、鉴定肿瘤驱动基因并钻研它们的有关信号通路、分子网络及调节机造对于改善癌症的检测、预防与医治阐扬着决定作用。利用遗传、生化、分子细胞生物以及职能检测的各类伎俩和细胞、幼鼠以及人体组织等各类钻研资料,我们多年致力于肿瘤基因的发现和职能、机造方面的钻研。先后发现了KAI1(抗癌1号)肿瘤转移抑造基因、KLF5和ZFHX3抑癌基因以及ERRF、U50、FOXA1、WWP1等多个肿瘤基因。我们还对KLF5和ZFHX3进行了较深刻的职能和机造方面的钻研,发现KLF5通过作为TGFβ信号通路的一个关键因子实显熹抑癌作用、KLF5乙;刺龆KLF5和TGFβ阐扬抑癌职能还是促癌职能、ZFHX3通过诱导细胞分化来实显熹抑癌职能、ZFHX3与激素信号相互作用并调节上皮细胞分化。将基因组和蛋白质组学伎俩利用到肿瘤资料将导致对肿瘤的各类生物学行为的深刻全面的相识,从底子上扭转对肿瘤的检测、预防、药物开发以及医治规划的造订。
汇报人简介:
Dr. Jintang Dong has expertise in cancer genetics and biology that is helpful for the success of this proposal. Both of his training and independent research have been on these areas. By publishing more than 100 research articles, he has the expertise in multiple areas of cancer biology research, including the discovery and characterization of cancer genes; functional evaluation of cancer genes in various models such as in vitro cultured cells, xenograft tumor models and genetically modified mice; understanding of how cancer genes function in both normal tissues and tumor cells; how genes are transcriptionally regulated; and how cancer genes regulate the balance of cell proliferation and differentiation in epithelial homeostasis. He has also been involved in head and cancer research at Emory, as he has been the Director for the Developmental Research Program in the Emory SPORE in Head and Neck Cancer. He has mentored multiple junior faculty members during his career so far.
